Microorganisms are usually colorless , so stains are used to make them visible  and help identify their shape, structure, and type . 📌 Learning Objectives After studying this topic, student will be able to: define staining explain purpose of staining describe types of staining techniques understand Gram staining steps identify microorganisms using staining methods 1️⃣ Definition of Staining Staining  is the process of applying color (dye)  to microorganisms to make them visible under microscope. Purpose: increase visibility identify shape differentiate types of bacteria observe cell structure 2️⃣ Purpose of Staining Staining helps to: identify bacteria study morphology differentiate gram positive and gram negative bacteria observe internal structures diagnose infection 🧠 Mnemonic – Purpose of staining "VISID" V – Visibility I – Identify bacteria S – Study structure I – Identify type D – Diagnose disease 3️⃣ Types of Staining Techniques Staining techniques mainly classified into: Simple staining Differential staining Special staining 4️⃣ Simple Staining Uses only one stain. Purpose: observe size observe shape observe arrangement Common dyes: methylene blue crystal violet safranin Example: Cocci appear blue under microscope. 5️⃣ Differential Staining Used to differentiate bacteria based on cell structure. A. Gram Staining (Most important) Gram staining differentiates bacteria into: Gram positive bacteria Colour: Purple Cell wall: Thick peptidoglycan layer Example: Staphylococcus Gram negative bacteria Colour: Pink Cell wall: Thin peptidoglycan layer Example: E.coli Steps of Gram Staining Apply crystal violet stain Add iodine solution Wash with alcohol Counter stain with safranin B. Acid Fast Staining (Ziehl-Neelsen stain) Used to identify bacteria with waxy cell wall. Example: Mycobacterium tuberculosis Result: Acid fast bacteria appear red. 6️⃣ Special Staining Techniques Used to identify specific structures. Capsule staining Shows capsule around bacteria. Example: Klebsiella Spore staining Shows bacterial spores. Example: Clostridium Flagella staining Shows bacterial flagella. Negative staining Background stained instead of bacteria.
